Madekwe was born in South London, England into an ethnically diverse family. Her father and mother are both English. She was a student at The Brit School when she was 14 years old. She went to The Royal Academy of Dramatic Art in London where she gained an BA in acting. While attending the academy, she performed in numerous stage productions such as the King Henry V playing Princess Katherine as well as Wuthering Heights portraying Catherine Earnshaw.

Madekwe was chosen to play Ashley Davenport for ABC's show The Beautiful Life. The two-episode series aired for just the duration of two weeks The CW and then it was cancelled. Ashley Davenport was her role in the ABC drama Revenge. After appearing on the show for the first two seasons the show was announced late May 2013 that she would leave the show after a guest appearance in the first episode of the third season. From 2014 until the year 2016, she appeared on Salem in the role of Tituba. Madekwe and her partner who has been with her for a long time, Iddo Goldenberg, were wedding in June of 2012.
